Well, Mario Kart and Wario Ware are naturally perceived as multiplayer games, but you can play either of them in single player, so although the games are more multiplay based, they aren't purely multiplay games. If that makes sense (which it does).
I'm sure the Mario 64 update will have single player features as well, although 4-player is nothing more than extremely interesting and exciting at this point in time. What exactly they're going to do with it I'm not sure, but you can guarentee that even if you don't have three mates, you're going to be able to enjoy this.
