We ground up a lettuce leaf and strained the juice through a piece of muslin. We divided this extract between 2 centrifuge tubes (moderately equally) and ran the two tubes through the centrifuge. We then obtained chloroplasts at the bottom of the tubes, we poured off the supernatant from both tubes and resuspended the pellets in two ways.

Tube A had 3cm cubed assay medium, while tube B had 2.7cm cubed distilled water and 0.3cm cubed assay medium. These were respun, the supernatants discarded again and then resuspended in 3cm assay medium.

Now the question i need help with is this.

If there was a significant difference in chlorophyll concentration (which there may or may not be) why would THIS difference not directly affect the rates [of DCPIP reduction]?) [20 Words]

I think it may be that there is sufficient chlorophyll concentration to allow the intitial reaction to start unabaited.
